Horse chestnut seeds contain aescin (β-escin), the most studied of their plant compounds. Aescin helps keep the walls of small blood vessels tight, so less fluid leaks into the surrounding tissue. That is why it is used to support healthy circulation and fluid balance around tissues, including in joint formulas.
There are no published studies in horses or dogs yet. The evidence comes from people: a Cochrane review of 17 randomised trials found that horse chestnut seed extract eased leg pain and swelling in chronic venous insufficiency, and lab studies on blood-vessel cells show how aescin works. L-Arginine
Circulation, hoof, and skin support

L-arginine is a functional amino acid involved in several important metabolic pathways. One of its key roles is serving as a precursor for nitric oxide, a signaling molecule that helps regulate blood flow and vascular function.

Astaxanthin is a naturally occurring carotenoid, usually sourced from microalgae, and is used in horses to support antioxidant protection and recovery, especially around training and competition. It helps protect cells from oxidative stress generated during exercise and can support healthy muscle recovery and overall resilience.
What makes astaxanthin stand out is its very strong antioxidant activity. In the scientific literature, astaxanthin has been reported to be more than 100 times stronger than vitamin E at protecting fats from oxidation (lipid peroxidation) in laboratory comparisons, and even higher for certain oxidative reactions, depending on the test method. In horses, recent peer-reviewed studies in racing horses found that astaxanthin supplementation was associated with reduced markers of oxidative stress and inflammation and improved antioxidant status, supporting its use as part of a performance and recovery plan. (nature.com)

Boswellia is used to support mobility and joint comfort in both dogs and horses because its boswellic acids help maintain a balanced inflammatory response, which can support more comfortable movement.
In dogs: Boswellia has been studied in dogs with osteoarthritis as part of multi-ingredient joint supplements. In two randomised, placebo-controlled trials, dogs on these supplements scored better on pain and mobility, as assessed by vets and owners. Because Boswellia was one of several ingredients, these studies can't show its effect on its own. It works best alongside a complete joint plan: healthy body weight, appropriate exercise and veterinary guidance. In horses: Boswellia is commonly used to support joints and post-exercise comfort, especially in older horses or horses in regular training. Research in horses is still limited. In a small placebo-controlled trial in 16 show jumping horses, a supplement combining Boswellia, turmeric and great mullein lowered the activity of pro-inflammatory genes in the blood after 10 days. As with dogs, it works best when the basics are right: a balanced ration, correct training, good hoof care and a sensible workload. Beghelli et al., 2023 (Life, MDPI)
